An animal components free cell dissociation solution, designed as an alternative to trypsin for the dissociation of mesenchymal stem cells (MSC) when used in conjunction with serum-free or serum-containing media
The use of animal-derived components in biopharmaceutical manufacturing is coming under ever-increasing regulatory scrutiny. Thus, there is a significant need to develop non-animal source products for cell culture handling.
Trypsin, an essential product for cell culture manipulation, is purified from animal-source materials with a notable chance of contamination with viruses, BSE, potential adventitious agents, and other undesired enzymes.
Biological industries developed MSC Dissociation Solution, a special non-animal alternative for porcine trypsin for the efficient detachment of mesenchymal stem cells.
This products was tested on two sources of hMSC (BM and AT) and found less damaging and more effective than other proteases.
To date, several commercial trypsin replacement solutions are available including, special non-enzymatic solution formulated with a proprietary mixture of chelators for gently dislodging adherent cell types from plastic. MSC Dissociation Solution helps maximizing the yield of functionally viable cells from culture vessels.
The use of plant derived enzyme is commonly used in cell isolation procedures where it has proven more efficient and less destructive than pancreatic enzymes on certain tissues including the dissociation of retinal neurons, (Shen J., et al., Japanese J of physiol. 1995), preparation of primary neurons from visual of rats (Huettner JE, Baughman RW J Neurosci. 1986), and for the isolation of smooth muscle cells (Kinoshita, K., et al., Am. J. Physiol. Gastrointest. Liver Physiol 2003 and Driska, S.P., et al., J. Appl. Physiol 1999).

Instruction for use:
Actual procedures and concentrations should be determined by experience with individual stem cell sources (see notes below).

Add 1ml of MSC Dissociation Solution to each T-25 tissue culture flask.

Incubate the flask at room temperature. Taping the flask's side can accelerate cell's detachment.

When cells are completely detached, quickly suspend the cells in 5-10 ml of growth medium.

Pellet the cells by centrifugation and discard the supernatant.

Suspend the cells in growth medium, and seed them as desired.
Notes

In order to avoid freeze and thaw cycles, it is recommended to divide the solution into small aliquots. After thawing, the solution is stable for 5 days at 2-8 °C.

The time needed to dislodge cells will vary depending upon stem cell sources and medium. If slower dissociation is desired, the MSC Dissociation Solution may be diluted with the Dilution Buffer for MSC Dissociation Solution (Cat No. 02-055-1).
Quality Control
MSC Dissociation Solution performance is tested using hMSC cells from bone marrow (BM) and adipose tissue (AT). Additional standard evaluations are pH, Osmolality and sterility tests. |
