Iscove's Modified Dulbecco's Medium (IMDM), with- L-Glutamine, without Alpha-Thioglycerol, without Beta-Mercaptoethanol, is a Dulbecco's modification and is considered a more complex and enriched media like Ham's F-12 and Connaught Medical Research Laboratories(CMRL-1066), among others, which are utilized not only for more specialized cell types but also as the basis for some of the more unique Serum-Free Media formulations.
Iscove's contains concentrations of Inorganic salts, Amino Acids (AA's) from L-Alanine to L-Valine and Vitamins from D-Biotin to Vitamin B-12 in addition to other ancillary constituents as D-Glucose (4.5g/l), Hepes, Sodium Bicarbonate, Phenol Red and Sodium Pyruvate. The original Iscove's formulation designed by none other than Iscove and associates, demonstrated that erythrocytes and macrophage precursor cells (e.g. primary Haemopoietic Cells) could be cultured in a totally defined serum-free medium when supplemented with albumin, lecithin, selenium and transferrin. IMDM is a modification of DMEM containing selenium, additional AA's, vitamins and other components.
Iscove's is a highly enriched synthetic media well-suited for rapidly proliferating and high-density cell cultures in a 5% CO2 atmosphere. IMDM reportedly not only supports Murine B Lymphocytes, Haemopoietic tissues from Bone Marrow, B-cells stimulated with Lipopolysaccharide, but also T-Lymphocytes and a wide variety of Hybrid cells. When utilized in conjunction with serum, it supports a broad spectrum of Mammalian cell types including but not exclusive to bone marrow, hybridomas and lymphocytes.
